项目名称:源于水生药用动物的多糖物质医药高效转化的关键技术及应用推广

项目简介:
多糖具有能量储存、药物递送及蛋白功能调控等多种医药功能。水生动物多糖更兼有陆生动物和水生植物多糖结构及功能优势,不仅水溶性好,功能团种类丰富、离子基团含量高,且抗原性小,无阮病毒等污染,已在医药应用领域呈现出多元化快速发展态势。然而,其“效率-质量-消耗-环保-安全”兼顾的转化工艺匮乏,“资源-结构-效能-制备-应用”间适配关系的调控及转化疲乏等资源高效利用的关键技术问题仍亟待解决,已成为制约行业发展和创新的关键瓶颈。
项目在8个国家自然科学基金及10余个省部级课题支持下,立足水生药用动物资源高效利用,瞄准其多糖医药转化及产品创新;围绕“转化工艺层面的可持续性,系统开发层面各环节间立体交叉关系的精准适配调控”这两大关键技术问题展开研究,实现水生药用动物多糖“集约制备-精准改性-构效适配-特异应用”间的协调统一。取得了如下创新性成果:
创新点1:建立以蛋白水解酶-凹凸棒石Pickering乳液为模板制备高酶活微球的酶高效固定技术,并以此构建超声同步耦合酶解的提取新方法,解决了传统热水提取的低效、高耗、质劣等技术瓶颈,使其提取时间缩短85%、溶剂减少40%、温度下降30%、提取率提高80%,实现“效率-质量-消耗”的高度集约化。
创新点2:创建基于循环冻融和特异吸附技术等的蛋白杂质绿色高效脱除新方法,突破了依赖Sevag等有机溶剂脱蛋白的技术壁垒,使有机试剂零使用,选择系数提高了0.9-4倍,实现“蛋白脱除-多糖稳定-环境保护”的协调统一。
创新点3:设计制备基于改良Hakomori法的甲基化自动反应装置,以此构筑“分步式”多糖甲基化方法,实现人员与碘甲烷零接触,使甲基化次数小于5次,攻克了多糖结构解析中甲基化这一关键环节的效率低、重复次数多、环境干扰大及操作风险高等关键技术瓶颈。
创新点4:制备得到田螺硫酸多糖及牡蛎多糖等12种新多糖,并根据多糖种属、特异结构及生物活性等,构建其在防治动脉粥样硬化、心脏洗脱支架和人工关节等5种应用体系。且针对多糖机械强度低、稳定性差及应用范围窄等性能及应用缺陷,基于曲线传质及自由体积等理论,构筑多糖-凹凸棒石新型复合功能材料,并构建其在缓控释辅料及药物稳定基质等4种应用体系。解决“资源-结构-效能-制备-应用”间适配不佳的关键技术问题。
项目已获授权发明专利26项,其中转化7项。关键技术已被江苏麦德森制药有限公司等8家企业推广应用于硫酸软骨素、虾青素等产品生产或研发,实现了相关产品高效、绿色、低碳及安全生产,形成了可食膜等多个特色鲜明的高附加值产品,取得了良好的社会和经济效益。近两年已累计实现新增产值1.8287亿元,新增利税4166万元,节支总额1488万元。发表论文53篇,累计引用1336次。
中国科学院院士林国强教授和第三方鉴定机构专家均评价认为该项目具有显著的创新性、先进性和实用性,在水生药用动物多糖医药转化的关键技术方面达到了国际先进水平。
